Repeated knocks to the head leads to newly recognized brain disease

ScienceDaily (Nov. 30, 2012) ? Reports on the danger of head trauma in athletes and soldiers has pervaded the news in recent years. NFL and NHL player deaths have catapulted this problem into the limelight, with stories appearing in the New York Times, NPR, ESPN, 60 Minutes and even television entertainment shows. Media attention has raced far ahead of the science on this degenerative condition. But scientists are catching on. They have given this disease its own definition -- chronic traumatic encephalopathy (CTE).

Different from traumatic brain injury, Alzheimer's, Parkinson's, or ALS, they say CTE can strike adult and youths alike. Special coverage by Alzforum, the leading news source on Alzheimer's and related diseases research, summarizes the status of research in this burgeoning field as researchers take steps to diagnose and treat CTE.

?CTE can start months or years after brain trauma has occurred, either in contact sports, military service, or perhaps even partner violence,? Admiral Regina Benjamin, the U.S. Surgeon General, recently told scientists and advocates at the first-ever CTE conference at the Cleveland Clinic Lou Ruvo Center for Brain Health in Las Vegas, Nevada. There, experts came together to define CTE, exchange the latest data, develop a diagnosis, and lay out a research plan (see Part 1).

Some of the newest data comes from boxers and other martial artists who have joined the Professional Fighters Brain Health Study in Las Vegas. The fighters? history as well as fight statistics are combined with yearly mental tests, brain scans, speech samples, and a blood draw. The aim is similar to studies in Alzheimer?s that have helped identify early signs of disease and track its progression. Scientists want to find out how concussions match up with brain damage and symptoms. Once this study ends, it may generate a tool to help determine when fighters should retire (see Part 2).

Additional information will come from DETECT, a biomarker study of football players, as well as a growing tissue bank of athletes? and military veterans? brains. Of 136 tissue donations, 100 brains have been analyzed and 80 had CTE hallmarks. They showed a unique pattern of brain change?expansion in its fluid-filled cavities and shrinkage of its medial temporal lobe?and extraordinary deposits of a toxic form of the tau protein that expand with age. From this data, scientists have begun to outline stages of CTE from beginning to end. It seems some athletes and soldiers who have been diagnosed with Alzheimer?s or Parkinson?s may have had CTE instead (see Part 3).

Other research suggests that in CTE, toxic proteins creep from the site of impact to further areas of the brain. Because toxicity moves between cells, not people, researchers refer to the spread as ?templating,? not ?infectious.? They are developing mouse models to help determine how this spread occurs (see Part 4).

Scientists are also drafting up the best way to diagnose the disease during a person?s life (see Part 5) and are pushing for measures to prevent CTE in children, who are more susceptible to developing the condition after head trauma (Part 6).?

Israelis question too, 'Who wants this war?'

Residents of southern Israel only reluctantly express support for a potential expansion of the conflict in Gaza and express sympathy for the suffering on the other side of the border.

Ashdod and Kiryat Malachi, Israel

Just days after Adi Pito married a woman from the town of Kiryat Malachi, he heard that a rocket from Gaza had struck an apartment building there, killing three residents.

?I was worried about my wife,? says Mr. Pito, who was in a nearby city at the time. ?If you ask me, I think we have to destroy Gaza. I think they are animals, not people,? he says, pulling at his new wedding ring as he stood outside the damaged apartment yesterday. ?It?s the right thing to do.?

But in towns across southern Israel that have been hit by rockets, other residents only reluctantly express support for a potential expansion of Israel?s Pillar of Defense operation and express sympathy for the suffering on the other side of the border, where at least 53 Palestinians have been killed and hundreds injured since the operation began five days ago.

?When people say, ?Let?s kill them,? I don?t think they really mean to do it,? says Yehudit Bar Hay, a trauma expert at the Israel Center for Victims of Terror and War, known as NATAL. ?It is an angry feeling but ? we don?t want to kill the people of Gaza. We see the mothers and the children, we are sorry for them.?

Even those who are ?very, very injured? from the trauma of living under rocket fire say they don?t want to hurt anybody, adds Ms. Bar Hay, who lives less than a mile from the Gaza border.?

Israeli Prime Minister Benjamin Netanyahu said today that Israel?s military ?is prepared for a significant expansion of its operations,? even as at least 16,000 reservists have been called up for a potential ground invasion. The Defense Ministry has authorized the calling up of as many as 75,000 reservists ? more than six times as many as participated in the 2008-09 Gaza war.

But while some Israelis say war is necessary or inevitable, few are happy about it.?

?Who wants this war? Nobody in Israel,? says Ashdod resident Bebert Avitan, his pink-rimmed glasses hanging from his neck.

Part of the reason for a lack of public pressure may be the effectiveness of Israel?s Iron Dome system, which has kept casualties very low despite a barrage of rockets from Gaza. See today's Monitor story on the Iron Dome system.?

?Hamas apparently has much greater capability than it had in the last war on Gaza,? says Galia Golan, a political scientist at the Interdisciplinary Center Herzliya, but notes that there hasn?t been a parallel uptick in Israeli casualties. ?Because Iron Dome has been relatively successful, there has been less pressure on the government or within the government to launch a ground attack.?

Only three Israelis, all from one building in Kiryat Malachi, have been killed in the recent escalation.

As the evening news shows a picture of a bombed out building, lifelong resident Masodi Sugaker says, ?It?s so sad to see this. To [have to] make everything new after this? Why? Because of Hamas.?

?Hamas exploits their own people, the Palestinians,? says she adds.

Her sister, Hanna Shukrun, adds, ?We don?t want [war],? because there are kids [in Gaza].

But she says, ?Israel can?t just sit here and do nothing?. Our army doesn?t need to wait until they have many weapons.?

(This story was edited after posting to correct the name of Galia Golan from IDC Herzliya. We apologize for the error).

A Friend for Health | Fitness & Wellness News

by Michelle Sutton-Kerchner

Many households own at least one pet. In addition to companionship and limitless love, these furry critters provide interesting health benefits ?

Opening your home to a pet is a large undertaking, one that requires extensive consideration. The time and financial commitment is ongoing for many years. Short of adopting an animal on his/her ?last leg,? you can expect a pet to be part of the family for many years. If considering this adventure in care and companionship, add improved health? yours? as one of the pros.

Exercise Opportunities

Canine friends, one of the most common pets, demand activity. Especially in puppy years, dogs need to burn off energy through frequent exercise. Their needs require them to go outdoors on occasion, which also gets their owners off the couch (sometimes out of bed) and heading for fresh air. Walking, ball tossing, and other doggie stunts require physical output from pets and their humans.

If looking for motivation to exercise, a dog is one of the best pets to own. A bunny or bird doesn?t require much movement on your part, unless they escape from their cage. In that case, any pet with the slightest Houdini skills can conjure up a workout for you.

Enjoy creative exercise opportunities.

The additional spurts of energy expelled during jaunts with a pet enhance your formal workouts. When weight loss goals plateau, adding 20 minutes of additional exercise during the day has proven to jumpstart a stalled loss. Having an active pet can require that extra output, revving metabolism and increasing stamina. You may find workouts are enhanced through a pet routine that involves regular activity.

Pets also offer a sense of belonging. For the shy or introverted, you are instantly part of an animal-loving community. Social by nature, dogs often need to greet passersby, especially those with other dogs. Think of them as a conversation starter with legs. You might be amazed at the friends introduced to you by your pooch.

Dog walking, which provides exercise and possible social interaction, may be the reason their owners have improved health over cat owners. Although any pet ownership showed a positive influence, dogs outweighed the perks of cats. Pet dogs? ability to facilitate social connections and increase physical activity may enhance physiological and psychological human health more directly, according to research published in the British Journal of Health Psychology.

Decrease the (Blood) Pressure

Where does one go to test the theory pets reduce stress and lower blood pressure? A group of New York City Stockbrokers, of course! A study assessed the effect of this form of social support in response to stress in 48 brokers being treated with medication for hypertension. Heart rate, blood pressure, and renin activity (an enzyme that increases with stress) were monitored. All participants? pre-treatment blood pressure readings were higher than 140/90 hg/ml. Participants were non-smokers, without other health issues, who lived alone.

Half the group added a pet dog or cat to their regimen. Their cardiovascular measures remained significantly more stable during stressful situations. Tense times are precisely when hypertension medication cannot help. Medications mainly control blood pressure under normal circumstances. Evidently, pets don?t have this fair-weather-friend reaction. Even during tough times, a pet provides calming support.

Many from the pet-less half of the study proceeded to find their own furry friend. Without a pet in their lives, blood pressure, heart rate, and renin levels almost increased to pre-treatment levels during stress, even while on medication. Bring in Rover!

Much of the lowered blood pressure outcome can be attributed to a pet?s ability to reduce stress. Soothing snuggles, consistently cheerful greetings (even if you forget to bring out the trash), and playful activities make for a tension-reducing lifestyle. Don?t forget the increased exercise common for pet owners, which reduces stress and offers countless mind/body perks.

A friend in need is a friend indeed.

Walter Reed Army Medical Center uses dogs to help soldiers deal with post-traumatic stress disorder. Veterans with pets are able to re-enter society more successfully. A pet decreases the threat of suicide in this population, which is one of the biggest health threats facing veterans. Beyond acting as a stress buster, pets offer love, companionship, and the ability to be needed? all without asking for the harsh details experienced during service time.

Lower Triglycerides and Cholesterol

According to the Centers for Disease Control pets can decrease these often troublesome blood levels. Some researchers found pet ownership lowers cholesterol by about five points. These heart-healthy contributions may be a result of the pet?s presence or the increased activity they bring. A pet?s unconditional love provides a sense of security and companionship when all else may be banished to the dog house.

Decrease risk of cardiac events.

Those at risk for cardiac disease may find a pet cat to be a lifesaver. Cat owners are 30 percent less likely to have a heart attack, and 40 percent less likely to suffer a cardiovascular incident like a stroke. Dogs have been known to assist their owner?s recovery from a serious illness, including heart attacks. Dog owners are eight times more likely to remain alive one year after having a heart attack than pet-less counterparts, according to Marty Becker, DVM, veterinary consultant and author.

Improve Other Chronic Health Issues

Dr. Becker believes pets can be the best medicine, especially when a person is dealing with chronic pain, such as migraines or arthritis. By reducing anxiety, which exacerbates pain, pet owners hurt less. Studies focused on acute pain found adults who used pet therapy required 50 percent less pain medication than those who did not.

Pets seem to connect on a level unreachable by most humans. Some epileptics are known to utilize dogs to help detect approaching seizures. Also, a study published in Diabetes Forecast, the American Diabetes Association?s magazine, found one-third of pets living with diabetics would change their behavior when their owner?s blood sugar level dropped. The pets included in the study were mostly dogs, but also included cats, rabbits, and birds.

Researchers attribute this perception to chemical changes in the owner?s body. The study inspired organizations like Dogs4Diabetics,?whose motto is ?Our Dogs Save Lives.? They train dogs to be companions of patients at risk for unstable blood glucose levels.

Expand your social circle.

Those suffering mild to moderate depression show improved mood through pet ownership. Taking care of a pet can help improve self-worth and increase social interaction. Indirectly, it also can help you take better care of yourself when this alone is a struggle. Being relied upon by another breathing being tends to reflect how much we truly can accomplish, in spite of our thoughts. And, simply petting animals is a soothing, repetitious act with meditative-like qualities.

The Motivation

Motivation to get in shape through pet ownership can begin even prior to acquiring a pet. Horse care can encompass all sorts of ?circuit training.? If you really want a workout, consider getting a pet horse. Just be sure to accomplish some intense functional training first or you could be left out to pasture by this four-legged friend.

Often, there is the ultimate motivation: a child?s begging eyes. One study found pets dramatically improve immunity. Children ages five to seven years attended three weeks more school annually than their pet-less pals. The holidays are approaching, along with New Year?s Resolutions to live a healthier life. A new pet can be the quintessential Christmas present.

Eurasianet.org spreads misinformation about the Armenian church in Nakhichevan

14:47, 28 November, 2012

YEREVAN, NOVEMBER 28, ARMENPRESS. The Armenian church built in the 19th century in Nakhichevan has not been merely turned into a cattle shed, but also was introduced as an "Albanian sacred place." The Chairman of a fund studying Armenian architecture Samvel Karapetyan referred to the misinformation published in Eurasianet.org website in his videoblog. As reports "Armenpress" Samvel Karapetyan stated that in the picture of the Eurasianet article is depicted St. Gevorg Church of Dashbulag village, which was entirely Armenian populated before 1918. The church was built in 1823, when "the Albanians" had left the stage 1000 years ago.

Samvel Karapetyan states: "Azerbaijan paid USD 4, 5 million to import cows from Germany and Austria, the photographer took the picture of these cows in Dashbulag, which was entirely Armenian populated before 1918. During Musafat administration the Turkish army massacred all the Armenians of the aforementioned village on their road to Baku. One can clearly see in the picture that the church became a cattle shed without doors. Under the picture there is an inscription saying that this is an Albanian church. According to Azerbaijani fabrication the Albanians are their ancestors. Hence, it becomes a monument constructed by the Turkish Christians, which is being treated as a cattle shed.

But the fact is that the church in Dashbulag was called after St. Gevorg, built in 1828. Among other things it is noteworthy that there had been records about the construction works. There is information about the aforesaid church in "the Albanian Country and its Neighbors" by Bishop Makar Barkhudaryants."

Gasoline Prices And Hybrid Cars | Gardening Gang - Lambert ...

The appeal and acceptance of numerous hybrid vehicles have grown particularly with the issues about high gasoline prices as nicely as a worsening air pollution. Here are some bits of helpful information that would support you understand far more about hybrid vehicles and how they might support you save on gas and be somehow protected from worries about increasing gasoline costs.

A hybrid automobile is a sort of auto that makes use of at least two different fuel sources to make it operate. Both fuel sources usually are getting used with each other in some situations to aid propel the car much more effectively. There are a number of various combinations possible for hybrid vehicles, but the most frequent hybrid car so far is the gas-electric hybrid.

The gas-electric hybrid auto, also known as the hybrid electric automobile or HEV, is recognized to make use of a gasoline internal combustion engine or ICE and a separate electric motor to assist power it. While the ICE makes use of gasoline to make it run, an electric battery is employed to retailer the electrical power that powers the hybrid vehicles electric motor. The HEV generally has a gasoline engine that is smaller than a standard 1 utilized in common gasoline powered automobiles. Use of far more advanced technologies tends to make this feasible and allows the HEV have better running effectiveness as properly as substantially decreased pollutant emissions.

Aside from the gasoline engine, the hybrid electric vehicle also has a special electric motor built in that not only assists supply added energy to the vehicle but also acts as a generator when not becoming employed. The electric motor can act as a generator in situations exactly where it is not becoming utilized to aid charge the battery for added efficiency.

In a common HEV set up, the car uses its electric motor when becoming propelled at very low speeds. The gasoline engine acts as a secondary power source when the HEV needs significantly necessary power as in the case of climbing up an incline. The gasoline engine also has the capacity to compensate the electric motor with power whenever the car wants it in order to go more rapidly. The gas and the electric motor can also perform in combo at certain instances if required. mil specifications

Due to the fact the hybrid electric automobile makes use of both an electric motor as effectively as a gasoline motor, a substantial improvement in auto mileage is somehow achieved. A hybrid electric automobile or HEV can run longer distances using the same amount of fuel as compared to a conventional gasoline powered auto. Whenever the electric motor is needed, gasoline use is avoided. This benefits in fewer gas used when running the exact same distance as a conventional gasoline powered automobile.

And simply because the hybrid electric auto has a smaller sized gasoline engine, the hybrid vehicle can also runs a lot more efficiently simply because of lesser engine weight as compared to the typical car. The working components of the hybrid automobile engine also are smaller and need less energy to move. This efficiency makes the hybrid electric auto rather a wonderful choice for individuals concerned with rising gasoline prices. Using the hybrid car can assist drivers conserve a substantial amount of gas when traveling. Not only that, using the hybrid auto can also help in decreasing pollutant emissions by using less gas whilst traveling.

After completing a grueling regular season, members of the MSU football team?s (6-6 overall, 3-5 Big Ten) junior class will use the next few weeks to decide if they?ll return to school next season or forgo their senior year to enter the NFL draft.

With head coach Mark Dantonio potentially having the most talented junior class in his tenure, highlighted by running back Le?Veon Bell, tight end Dion Sims, defensive end Will Gholston and cornerback Darqueze Dennard, among others, there could be a number of players deciding their future in the coming weeks.

?If they?re going to stay, they?ve got to have good reasons to stay, otherwise they?re not happy. If they?re going to leave, they?ve got to have good reasons to leave, otherwise they?re not happy,? Dantonio said.

?The first thing that you would do as a young player is you could petition the NFL to get a read on what possible draft pick you would be, keeping in mind that ? 47 percent of the people that say, ?I?m going to go,? they never get drafted.

?I?m going to make sure I?m not selfish because I?ve never tried to be that for our players, I?ve tried to be for them. So that?s how we handle it.?

Top Signs of Foundation Trouble | Home and Garden

Homeowners know that a home is a major investment and that taking care of the home is very important not only to protect that investment but also to ensure a safe place for you and your family to live in. One of the most important aspects of taking care of any home is making sure that the home rests on a solid foundation. As such, it is important to be aware of possible foundation problems and to ensure that you monitor for problems and call in a professional as soon as you see a possible issue.

Signs of Foundation Trouble

Sometimes, a foundation problem is obvious such as when a large crack develops in the walls of your basement. However, there can also be other signs that your foundation is having issues that may not be as obvious. Knowing what to look for is important, and here are a few signs of trouble with your foundation that you should be aware of and pay attention to:

1. Sloping or uneven floors in the basement. This can be a sign that the walls and basement floor are starting to separate or pull away from each other. It is a serious issue and one you should deal with promptly by calling in a foundation repair expert.

2. Cracks or shifts in your crown molding. Crown molding is molding between your sealing and your wall. If your crown molding begins to pull away at the sides (i.e. in corners or where two pieces meet) this is a sign that your home is shifting and your foundation might be in trouble. This is especially true if the crack begins in the wall and continues up into the molding.

3. Cracks or gaps between the walls and ceiling. If your home does not have crown molding, then you may notice gaps developing between the wall and ceiling. This, too, is a sign that the foundation of your home is shifting and that your foundation needs attention.

4. Cracks on your home?s exterior in the brickwork, or the walls separating form your home. This is an extremely serious sign of foundation problems and it means that you must contact an expert right away to preserve the structural integrity of your home.

5. Separation at the windows, doors or garage door. Essentially, like most of the other signs, shifting and pulling away of parts of your home that used to be tight are all indicators that your home?s foundation is beginning to develop problems.

6. Inability to tightly or properly close windows and doors. If your foundation begins to sink, door frames may begin to sag and you may be unable to properly open or close doors. Likewise, a sagging or shifting foundation can cause your windows and doors to become out-of-square and more difficult to open or close.

If you begin to notice any of these issues with your foundation, it is very important that you contact an expert. The longer you wait, the worse the problems in your home may become and the more expensive it may be to fix all of the issues that have developed due to your sagging, shifting or damaged foundation. Don?t hesitate to take action. With the right expert, you may be able to find a simple and easy solution to your foundation issues.

While his mother had been Winston Churchill?s private secretary, business is also in the blood: He is the son of a businessman who traded in whiskey during America?s prohibition years and later worked for a company that survived the ban by selling communion wine.

Born in London in 1956, Justin Portal Welby studied law and history at Trinity College, in Cambridge, England, after the initial schooling that made him an "Old Etonian."

A career in the oil industry beckoned and he spent 11 years in the sector as a senior executive, based in Paris and London. He worked on projects in the North Sea and the Niger Delta, gaining a familiarity that would come in useful in later years as a cleric traveling to the West African reaches of the Anglican Communion, Nigeria.

In 1983, his seven-month-old, first-born daughter was killed in a car crash, leading to "a very dark time" for him and his wife. But it also "bought us closer to God," he has said.

He left behind his career in business in 1987 to train an Anglican priest, later telling business magazine Money Marketing "I was unable to get away from a sense of God calling."

He became a deacon in 1992 after taking a degree in theology, serving later as a curate in the Coventry diocese and was made a rector in 1995 before being made a canon at Coventry Cathedral in 2002.

At Coventry, he was involved in international conflict resolution before becoming dean of Liverpool in 2007. He was elevated to the fourth most senior post in the Church of England in November 2011, when he became the bishop of Durham.

Thousands protest after Bangladesh factory fire

Thousands of textile workers gathered in Bangladesh in protest factory conditions following a massive fire that killed 112 people. NBCNews.com's Dara Brown reports.

By NBC News wire services

DHAKA, Bangladesh --?Thousands of Bangladeshi workers blocked the streets of a Dhaka suburb Monday, throwing stones at factories and smashing vehicles, as they demanded justice for at least 112 people killed in a garment-factory fire that highlighted unsafe conditions in an industry rushing to produce for major retailers around the world.

Another fire broke out in a multi-story garment factory in a Dhaka suburb on Monday, but a fire department official said the blaze was under control and there were no immediate reports that anyone had died in the latest blaze.

Some 200 factories were closed for the day after the protest erupted in Savar, the industrial zone where Saturday's deadly fire occurred. Protesters blocked a major highway.

Workers shout slogans Monday as they protest against the death of their colleagues after a weekend fire in a garment factory in Savar, Bangladesh, killed more than 100 people.

The government announced that Tuesday will be a day of national mourning, with the national flag flying at half-mast in honor of the dead.

Fire official: No emergency exit
Investigators suspect that a short circuit caused the fire, said Maj. Mohammad Mahbub, fire department operations director. But he said it was not the fire itself but the lack of safety measures in the eight-story building that made it so deadly.

Fire sweeps clothing factory in Bangladesh -- more than 100 killed

"Had there been at least one emergency exit through outside the factory, the casualties would have been much lower," Mahbub said.

He said firefighters recovered at least 100 bodies from the factory, and 12 more people died at hospitals after jumping from the building to escape the fire.

Local media reported that up to 124 people were killed.

"I haven't been able to find my mother," one worker, who gave her name as Shahida, told Reuters. "I demand justice. I demand that the owner be arrested."

Mohammad Ripu, a survivor, said Monday that he tried to run out of the building when the fire alarm rang but was stopped.

"Managers told us, 'Nothing happened. The fire alarm had just gone out of order. Go back to work,'" Ripu said. "But we quickly understood that there was a fire. As we again ran for the exit point we found it locked from outside, and it was too late."

Ripu said he jumped from a second-floor window and suffered minor injuries.

Mahbub said the fire broke out on the ground floor, which was used as a warehouse, and spread quickly to the upper floors. He said many workers who retreated to the roof were rescued, but dozens of others were trapped; firefighters recovered 69 bodies from the second floor alone.

A fire blew through an eight-story clothing factory in Dhaka, the capital of Bangladesh on Saturday night. The factory made products for Walmart and other U.S. companies. NBC's Kate Snow has more.

Many victims were burned beyond recognition. The bodies were laid out in rows at a school nearby. Many of them were handed over to families; unclaimed victims were taken to Dhaka Medical College for identification.

Hazardous conditions are widespread
The garment-factory fire was Bangladesh's deadliest in recent memory, but such dangers have long been a fact of life as the industry has mushroomed to meet demand from major retailers around the world.

At least 500 people have died in clothing factory accidents in Bangladesh since 2006, according to fire department officials.

The Savar factory is owned by Tazreen Fashions Ltd., a subsidiary of the Tuba Group. Neither Tazreen nor Tuba Group officials could be reached for comment.

The Tuba Group is a major Bangladeshi garment exporter whose clients include Wal-Mart, Carrefour and IKEA, according to its website.

The Bangladesh Garment Manufacturers and Exporters Association said it would stand by the victims' families and offered $1,250 to each of the families of the dead. The association's acting president, Siddiqur Rahman, said on a talk show late Sunday that Tazreen's owner was to meet with group representatives on Monday.

"We will discuss what other things we can do for the families of the dead," Rahman said on Rtv, a private television station. "We are worried about what has happened. We hope to discuss everything in detail in that meeting."

Cell biology: Calcium 'accelerator' keeps cell power supply going

ScienceDaily (Nov. 25, 2012) ? A team of scientists from Temple University School of Medicine and the University of Pennsylvania has moved another step closer to solving a decades-long mystery of how the all-important flow of calcium into the cell's power source, the mitochondria, is controlled.

By painstakingly shutting down the activity of 50 genes, one at a time, they have identified a protein, MCUR1, which hugs the inside of the mitochondrial membrane and is part of an elaborate mitochondrial channel pore system. MCUR1 acts as an accelerator to help regulate calcium coming into the mitochondria from the cell's large reservoir.

The results, appearing November 25, 2012 in an advance online issue of the journal Nature Cell Biology, may also point to new treatment opportunities. Understanding how to manipulate MCUR1 may help in the development of treatments for disease conditions involving excessive calcium in the cell, such as cardiovascular disease and stroke.

"Calcium is a key to regulate many fundamental processes in cells," said co-senior author Muniswamy Madesh, PhD, Assistant Professor of Biochemistry at Temple University School of Medicine and a member of Temple's Center for Translational Medicine. "Excessive calcium in the cell's mitochondria could lead to heart and neuronal mitochondrial dysfunction and cell death. This pathway could be contributing to disease conditions during ischemia/reperfusion injury and stroke, and this discovery opens up possible therapeutic interventions."

Maintaining calcium at an appropriate level is crucial for cells to work properly, and especially important in the mitochondria. Cells rely on mitochondria to generate usable energy in the form of the chemical ATP, which is necessary to carry out normal cellular and metabolic activities. ATP production in turn depends on calcium -- more specifically, charged calcium ions -- that can enter into the mitochondria from the cell's vast supply in the cytoplasm. Dr. Madesh, co-senior author Kevin Foskett, PhD, at the University of Pennsylvania, and their co-investigators recently described the role of a gatekeeper protein in maintaining a calcium "set point" under normal resting conditions in the mitochondria. But a long unanswered question remained: the details of how calcium entry into the mitochondria is controlled.

How MCUR1 Dictates Calcium Load

In the current study, Drs. Madesh and Foskett and their co-workers may have come closer to solving this puzzle. Dr. Madesh and his group sought to identify the genes involved in the flow of calcium into the mitochondria. They developed a way to use a technology called targeted RNA interference (RNAi) to screen 50 mitochondrial proteins, systematically testing whether eliminating the function of each of these genes individually altered the movement of calcium into the mitochondria. They found a mitochondrial inner membrane protein -- MCUR1 -- that regulates a calcium channel pore during active calcium uptake.

MCUR1 is part of a calcium channel pore called the uniporter, the existence of which has been known for some five decades. Recent studies identified two important pieces of the pore -- a subunit protein, MCU, and the gatekeeper protein, MICU1, to maintain calcium levels at a resting, set point. The researchers found that MCUR1 interacts with MCU to accelerate the movement of calcium into mitochondria when the cell's calcium level rapidly rises.

"MCUR1 is an essential third component of the uniporter complex," Dr. Madesh said. "In the absence of MCUR1, mitochondrial calcium uptake is markedly reduced, with adverse cellular consequences, including compromised cellular bioenergetics."

Without this accelerator, the mitochondria channel pore alone cannot take up calcium. When MCUR1 is physically attached to the pore, it is functional, and when it is not attached, it is much less active. "The regulator is always on, and its activity level increases when the extra-mitochondrial calcium levels increase. When there is a high calcium level, say during a disease state, the MCUR1 senses this and facilitates the channel activity, dumping calcium inside," Dr. Madesh explained.

Calcium Control as a Targeted Therapeutic

Dr. Madesh explained that in some disease conditions -- such as ischemic reperfusion injury (tissue damage that occurs when blood returns to tissue after a period without oxygen), stroke involving brain injury from ischemia, and myocardial infarction -- calcium floods into the mitochondria. "The mitochondria have a massive membrane potential and grab external calcium. The mitochondria then are overloaded and rupture, leading to cell death and possible organ injury."

Because some diseases are characterized by an overabundance of calcium in the mitochondria, a possible therapy could involve slowing down the amount of calcium coming into the mitochondria. "We want to control the calcium entering the mitochondria," he said.

"Further studies already underway should reveal the precise molecular mechanisms by which MCUR1 accelerates the channel pore MCU, providing insights into mitochondrial bioenergetics," said first author Karthik Mallilankaraman, PhD, a postdoctoral fellow in the Department of Biochemistry and the Center for Translational Medicine at Temple University School of Medicine.

UC Finance urges contractors against paying unnecessary taxes

11/20/2012 (press release: DPRi) // Helen Reid

UC Finance has called upon Britain?s contractors to celebrate National Freelancers Day by taking steps to check whether they are one of the independent workers currently paying ?1billion in unnecessary taxes every year.

While controversy continues to rage around the questionable tax contributions of many large companies, UC Finance?s research suggests that half of the UK?s estimated 200,000 contractors could be losing up to 11% of their annual earnings after being forced to work through an Umbrella Company by HMRC?s IR35 regulations.

?Given the news that has emerged recently to suggest that companies like Starbucks and Amazon are paying only a fraction of the taxes the exchequer might have expected to receive, it is ironic that around the country, individual contractors and freelancers are paying over the odds due to the confusion surrounding the IR35 rules,? said UC Finance Managing Director Joanna Holloway.

?The good news for freelancers is that there is more support for them than ever before from within the industry, with free advice and online tools available to help determine whether IR35 applies to them and a variety of products and services dedicated to providing legal protection and financial support.?

Many existing contractors are currently working through Umbrella companies due to fears that their independent status could be questioned by HMRC under the IR35 regulations. It?s an expensive option, with Timesheet Finance provider UC Finance calculating that a freelancer earning ?350 a day could be entitled to save ?10,000 a year - including all costs - by setting themselves up as a one man limited company.

With recent statistics from Freelance Advisor suggesting that more than half of the UK?s nine to five workers considering going it alone, UC Finance has joined forces with leading industry experts to provide existing or potential freelancers with advice, information, insurance, accountancy and finance to bridge the gap and support the ever increasing entrepreneurial spirit currently alive and kicking in the UK today.

In order to create a hassle free escape route for freelancers currently working through Umbrella Companies, and workers considering going it alone, the company has partnered with business contracts and assurance specialist Qdos and contractor accountancy firm FW Accounting to create a cost effective one-stop solution.

After using the campaign?s free online calculator to check whether leaving the Umbrella Company setup would save a significant sum, contractors have the option of contacting UC Finance directly by telephone, email or live chat to discuss the process of switching to a limited company format. The campaign?s accountancy partners will provide free business incorporation as part of the solution, with Qdos supplying the necessary business insurances and UC Finance stepping in to provide the financial support required to keep the participant?s income stream flowing steadily throughout the process.

Holloway continued: ?Given the increasing numbers of individuals entering the freelance market due to the current economic climate, it is unacceptable that Freelancers and contractors across the UK are losing significant sums of money because they?ve been sold or frightened into taking an option that simply isn?t the best solution for them. Recent years have seen ?freelancer bashing? become a national sport - it?s high time they were given a (tax) break.?

About UC Finance

UC Finance was founded to bring a refreshingly ethical, straightforward and transparent approach to the financial sector. Emerging with a bright new ethos from the wreckage of the traditional banking industry, the company provides freelancers and contractors with the essential low cost borrowing facilities frequently denied them by High Street banks.

Combining a 21st Century approach to the Finance sector with old world values, the company treats self-employed customers with the respect they deserve, offering accessible finance packages to a range of one-person limited companies.

All costs, which are fully tax deductible, are agreed up front with no hidden charges. Clients pay less than 4% of their invoice value, on average, for the full service : - cash advanced, invoices raised, and collections undertaken.

Ranbaxy recalls generic Lipitor in U.S., shares fall

MUMBAI (Reuters) - Indian drugmaker Ranbaxy Laboratories Ltd has voluntarily recalled its cholesterol-lowering drug atorvastatin in the United States, a development that could dent its image further in the world's biggest prescription drug market where the company faced an earlier ban.

Ranbaxy did not specify the reason for the recall of the drug, which largely drove the company's sales in the first of half the year. Shares of the company fell more than 4 percent following the announcement.

The recall of atorvastatin, a generic version of Pfizer Inc's Lipitor, will temporarily disrupt supplies in the U.S. market while the company conducts an investigation that is expected to take two weeks, it said. Lipitor is considered as the world's best-selling drug ever and at its peak generated $13 billion in one year for Pfizer.

Ranbaxy was the first company to launch generic Lipitor in the United States after Pfizer's patent expired last year. The Indian company enjoyed marketing exclusivity for the first six months that generics were allowed to be sold along with U.S.-based Watson Pharmaceuticals Inc.

"The development will impact the company's credibility to an extent," said Bhagwan Singh Chaudhary, a research associate at the brokerage IndiaNivesh. "There have been issues in the past (about compliance) and a recall suggests, corrective measures suggested by the U.S. FDA are not being implemented."

Generic Lipitor generated nearly $600 million in sales for Ranbaxy during its exclusive marketing period, Chaudhary said.

Other drugmakers including Indian rival Dr. Reddy's Laboratories Ltd have since launched their own generic atorvastatin versions in the United States.

Controlled by Japan's Daiichi Sankyo Co, Ranbaxy has had a troubled history in the United States where shipments from some of its manufacturing plants were banned from 2009 over a compliance-related probe.

The ban was troubling for Daiichi Sankyo, which acquired a majority stake in the Indian company for $4.2 billion in 2008.

Early this year, Ranbaxy agreed to make broader changes at its plants and agreed to appoint an external auditor to resolve the dispute with the U.S. Food and Drug Administration.

It has also set aside $500 million towards any settlement charges it might have to pay.

Shares of Ranbaxy, which is valued at $3.93 billion, were down 3.5 percent at 494.85 rupees as of 0950 GMT, underperforming a 0.07 percent fall in the Mumbai market.

Malaysia And Cambodia: 115. Baba Nyonya Museum

The Baba Nyonya Heritage Museum here is a must see. There are two ladies who do guided tours (the slimmer lady was really fun) and tours run for about 30-40 minutes. The museum is right in the middle of Chinatown and runs parallel to Jonker Street

About the Baba Nyonya Museum
The house was built in 1896, and was the family home of the Baba Chan family known as Rumah Abu. Chan Kim Lay, aged 83, is the fourth generation of his family.
It is in a well-preserved traditional Peranakan town house, built in 1896 by millionaire rubber planter Chan Cheng Siew. Today it is owned by William Chan and his family, who conduct tours of their ancestral home. The interior is that of a typical 19th-century residence and all the rooms are still as they would have been 100 years ago. The house contains family heirlooms and antiques, including Nyonyaware porcelain and blackwood furniture with marble or mother-of-pearl inlay, and silverware. There is also a collection of traditional wedding costumes, photographs and kitchen utensils. The kitchen sink has the name of William Chan's great grandfather carved on it. The information-packed tours are run regularly throughout the day.

Sunday 21st March
Today's "sightseeing" was on the Chinatown side of the Malacca River. I asked Liz (part owner of Heeren House) the best route to take to avoid backtracking and the Baba Nyonya Heritage Museum was first on my "list" seeing as it was in Heeren Street, only just up the road from where I was staying. The metal security door was locked and you had to press the bell. A lady came, asked did I want to do a tour, the next one was starting in about 15 minutes and let me in. Price was a few ringitts. She said no photos were allowed inside the museum (you can take as many pics as you like of the outside though) This (she said) was because people came, took photos and made copies of the furnishings. At the end of the tour you can buy souvenirs. (I bought postcards and took photos of the postcards.)

Above: Baba Nyonya Heritage Museum
The Chinese signboard above the main door way is called a Jee Ho.
We learnt the fascinating history of the Baba Nyonyas who are descendants of the Chinese who came to Malaysia and married Malays. The offspring were the Baba (male) Nyonya (female). They are also known as ?Straits-born Chinese? or ?Peranakans?. The Straits Chinese were of noble descent and adopted much of the Malay culture as their own. Elderly Chinese ladies were very conservative and would peek through the holes in the fancy carved timber to see who was in the room where the master of the house conducted his business affairs. Above: Baba Nyonya Entrance
As we walked through the middle door, we learnt that the present owner who is the grandson (or the great grandson?) of the original owner had turned his family home into a museum some years ago and was about 70 or 80 years old, and though he might be "old" his brain is as sharp as a tack. The Chinese man who originally bought the house, bought one house first, then the second then the third and turned them into one house. Originally the entrance was the door of the house to the right but that has now been locked and the middle house has the entrance. Above: The First Lady of the House
After walking in, we moved to the room on the right which is where business was conducted. Chinese ladies never took part in this and the wives of the business men sat in another room and played card games. The table had drink holders which could be pulled out from the table to hold cups/glasses. They must have been very bored playing cards and eating sweetmeats. A far different lifestyle than today.

Moving back into the centre and walking towards the back is a very large area with a sunken tiled area. There is no roof - houses were very narrow, had windows at the front only, so natural light was needed and was therefore necessary to have this area for when it rained, otherwise the house would have been flooded. Another interesting fact - the only door is at the front - there are no back doors. So bad guys had no way of entering the house. That is also the reason windows are at the front of the house only.

Moving toward the front of the house, just in front of the atrium is a beautiful staircase with carpet on the upper level. We were told to remove our shoes. The first part is circular and at the top on the right is a long hallway with sideboards displaying antiques. One of these held very, very old bottles of brandy and cognac. We were instructed not to sample them. Asking the tour guide if the taste would be better now due to age, she laughed and said she wasn't sure but nobody was game enough to try!

CCTV
We may think we invented CCTV, but those Chinese invented it first! They were very smart - after climbing up the stairs, we were led to the area in the middle of the house near the front and instructed to look down. There was a small square about 6" (15 cms), our guide pulled something and lifted it off and there we had a view of the entrance door below. She explained this was how the master of the house determined who he would allow to come into his home - if he didn't like the look of you, a bowl of water would be tipped out over you, so always hold an umbrella over your head!

Bridal Chamber

Above: Bridal Chamber
The bridal chamber which you see above is a most magnificant bed with rich tapestries and furnishings. This is where the master of the house and his bride slept. There is a ceremony held first before they jump into bed. As marriages were arranged, the bride had only seen her bridegroom's face through the peephole before the wedding, the groom hadn't seen the bride at all and they needed to get to know each other first. Such a sensible idea really.

There was a large mirror a few feet from the bed, the bride sat on a chair near the bed and the groom would look in the mnirror to see the face of his bride. This way he was prepared should he be unlucky enough to have married an ugly lady! (As I mentioned, such a sensible idea don't you think?) After this, they would then sit at a table and partake of drinks and foods while they became comfortable with each other. The bed is quite small by today's standards as people were quite a bit shorter and smaller back in those days.

Above: Inside the museum
Coming out from the bridal chamber we moved to the left along a passageway to the children's rooms. This passage had windows which faced the front and we could see the courtyard below. There was a staircase off to the right and for safety and security there was a folding door which opened at the top staircase area and unfolded to seal off access. Very good if the husband had had a night on the town and wanted to sneak in. Also very useful if the lady of the house was annoyed with him - all she had to door was seal this off and the poor old husband had to spend the night outdoors!?

Coming back down the stairs, putting our shoes back on we were led to the kitchens. Those poor servants had no modern day conveniences of today and must have been a very healthy bunch indeed. The master and his family ate at one table - there was another table (not nearly as glorious) where the servants ate. Beautiful china was arrayed and the more intricate the design, the wealthier the family. Blue and white china is used for funerals only. You can see in the above collage, top left hand photo,? the pink floral china which was in use.

The owner of the house had had installed modern toilets near here which we could use, and souvenirs were in a smallish alcove. It was a most instructive tour and should I find myself in Malacca again, I'd definitely do the tour again.

Linux Foundation vet explains setbacks in getting a Secure Boot key for Windows 8 PCs

Windows 8 dual-boot

Linux fans wondering why they still don't have a friendly UEFI Secure Boot option for Windows 8 PCs won't get a solution in hand this week, but they'll at least get an explanation. The Linux Foundation's primary backer for the alternative OS efforts, Parallels' server CTO James Bottomley, has revealed that Microsoft's requirements for signed, Secure Boot-ready code are tough if developers aren't entirely onboard its train of thought. The Redmond crew demands a paper contract signature (remember those?), agreements on work beyond the relevant software and a packaging process that complicates attempts to use open-source tools. Bottomley has already overcome most of these challenges, although he's still waiting for a Linux Foundation-specific key that should theoretically clear a major hurdle. Whether or not that leads to a remedy in days or weeks is up to Microsoft; in the meantime, we'll take comfort in knowing that a signature is so far a convenience for booting into Linux, rather than a necessity.

Locked-out Lundqvist finally finds game to play

(AP) ? Henrik Lundqvist finally found a game to play.

It won't be under the bright lights of Madison Square Garden, or in front of a hostile crowd in Philadelphia trying to rattle the man in the red, white and blue mask. But it will be meaningful and rewarding in a unique way.

Like players and fans all across North America and the world, the New York Rangers goalie misses the NHL. Lundqvist has run the gamut of emotions as the long lockout lingers. His thirst to get back on the ice will be quenched Saturday night ? however briefly ? as he joins other hockey stars in giving back to people who were ravaged by Hurricane Sandy.

Lundqvist will step on the ice in storm-damaged Atlantic City, N.J., along with Rangers teammates and foes from the Flyers and other teams to lend a helping hand to those recovering from the devastation. It won't have the intensity of a regular NHL game, but it will be filled with emotion and enjoyment.

"Absolutely. It's not only the hockey fans. You want to pay back to people working in the community around New York and people that got hit by this storm," said Lundqvist, last season's Vezina Trophy winner as the NHL's top goalie. "I was not there, but obviously I have a lot of friends who were still there, and I had people move into my place. ...

"I hope it's going to be a positive day, and that people who come in to watch the game get some energy and give the people some hope who need it."

"Operation Hat Trick" will be held at famed Boardwalk Hall on Saturday. All proceeds from the event will go directly to the Empire State Relief Fund, the New Jersey Hurricane Relief Fund and the American Red Cross, which is aiding New York and New Jersey families affected by Hurricane Sandy.

Brad Richards of the Rangers joined Flyers forward Scott Hartnell and partnered with Caesars Atlantic City to stage the event. Richards and Hartnell will serve as captains of the two teams that will feature other NHLers such as Martin Brodeur, Bobby Ryan, Steven Stamkos, Kimmo Timonen, Braydon Coburn, James van Riemsdyk and Simon Gagne.

As of Monday, 28 players had signed up to play.

"We are all overwhelmed by the damage and destruction caused to our area by Hurricane Sandy, and as professional hockey players we wanted to come together to help those who were victimized," Richards said.

The mere idea that Rangers, Flyers and other rivals could come together for a common goal is special in itself.

"To have the opportunity to be able to raise money for the relief effort by participating in this game is an honor," Hartnell said.

Lundqvist has been spending time at home in Sweden. His Thanksgiving plans included an international flight back to his hockey home that hasn't had hockey since the Los Angeles Kings captured the Stanley Cup in June.

This trip should be well worth it for the seven-year NHL veteran.

"I'm trying to stay busy," said the 30-year Lundqvist, who joined the Rangers in 2005 after an earlier lockout forced the cancellation of the previous season. "I've been skating every week, trying to stay ready in case something happens. I'm not sure if things are going to open up in the NHL or here in Sweden. Right now I'm just trying to stay ready, but it's tough, though.

"Just a couple of nights skating. It's not ideal, obviously, but it is what it is."

A glimmer of optimism that arose this week when the NHL and the players' association got together twice for negotiations was dashed on Wednesday when the league quickly rejected an offer it said it was eagerly awaiting. The NHL has already canceled all games through November and the highly popular outdoor Winter Classic. All-Star weekend is very close to being lost along with another round of games.

"I'm kind of tired of hearing that they're getting together," he said. "We need progress here. I don't know what to say now. The first couple of weeks you hope it's going to end fast, and now you've had two months of talking.

"It doesn't really matter. We just have to figure it out here with the owners and come to a solution. Everybody is suffering, from the fans, the players, people working with the game. It's not necessary. The game is strong enough that we should be able to figure it out."

Lundqvist has rolled with the ups and downs of the lockout that began in September, when the previous collective bargaining agreement between the players' association and the NHL expired. He hasn't signed up to play in Europe or anywhere else, but if a deal isn't reached soon, he might have to change his plans.

He just never thought the lockout would be going on this long.

"It's important that we try to give back to different communities and to the fans, but to us players, we miss the game a lot. We do," Lundqvist said. "This is our lives. There is a business side to this sport; there is no way around it. When it comes down to just playing the game, I miss the lifestyle, I miss traveling and playing the games and having the adrenaline.

"It's just such a big part of my life. It's definitely been a weird two months here not having that. That's why it's a lot of fun to get together and play the game."

The NHL generated record revenues of $3.3 billion last season when Lundqvist and the Rangers reached the Eastern Conference finals. Instead of being back on the ice to shoot for another run at the Stanley Cup, the star goalie is in a holding pattern he can't escape.

"I have all the emotions," Lundqvist said. "I have some anger, disappointment, some embarrassment, too. I understand that the fans are disappointed, and it's hard to explain the whole situation. I don't think a lot of people know all the details and how much we really sacrificed to try to come closer to the owners. It doesn't really matter. They deserve the game out there to be played right now. I definitely feel a little embarrassment that we can't figure it out sooner."
